Screening Platform

Description

The screening platform provides expertise for the development, miniaturization and validation of biochemical and cellular assays to be used for screening of small molecules libraries. Our instrumentation allows the setup of flexible, partially automated protocols, using different readouts including absorbance, luminescence, fluorescence, TR-FRET and cell imaging. The laboratory is equipped with a high-content screening (HCS) system combined with software for image analysis and data analysis. The platform supports Drug Discovery projects by performing both primary screenings as well as dose-response validation assays, orthogonal and secondary assays.

Equipments

Operetta-CLS (Perkin Elmer): High-content image system equipped with eight LEDs, for fluorescence (confocal and widefield) and transmitted light acquisition, air and immersion (water) objectives and Harmony image analysis software.

Glomax Discover (Promega) Microplate reader  for luminescence, fluorescence, UV-visible absorbance, BRET and FRET detection.

Spark (Tecan) multimode microplate reader equipped with a high-speed monochromator, dedicated photomultiplier for luminescence detection and hybrid technology (combination of filters and monochromators) for fluorescence readout.

In-Hood Bravo (Agilent) for automated liquid handling under sterile conditions. Equipped with 96ST and 384ST warheads.

Incucyte S3 (Sartorius): Live-Cell Imaging and Analysis System: Two-color Green/Red fluorescence plus HD phase contrast imaging. Equipped with 4X, 10X, 20X objectives on an automated turret. Support for three interchangeable vessel trays and over 700 vessels, compatible with flask, dishes, slides and microplates – up to six microplates in parallel.
